چکیده

With key roles in essential brain functions ranging from the basics of excitatory neurotransmission to the complexities of learning and memory, the N-methyl-D-aspartate (NMDA) receptor can be considered one of the fundamental neurotransmitter receptors in the brain. Named for its most potent exogenous agonist, the NMDA receptor has been thoroughly characterized via electrophysiological and pharmacological techniques, aswell as throughmolecular manipulations and transgenic knockout strategies. The NMDA receptor belongs to a family of ionotropic receptors for the excitatory amino acid glutamate and is characterized by high affinity for glutamate, a high unitary conductance, high calcium permeability, and a voltage-dependent block by magnesium ions. this article focuses on the basic biophysical properties and physiological functions of NMDA receptors and how these are modulated by various signaling molecules and biochemical cascades under physiological conditions.

The antinociceptive effect of 17β-estradiol in the nucleus paragigantocellularis lateralis of male rats may be mediated by the NMDA receptors

Introduction: The nucleus paragigantocellularis lateralis (LPGi) is involved in the descending pain modulation. The neurostreoid, 17β-estradiol found in the PGi nucleus and modulates nociception by binding to estrogen receptors and also by allosteric interaction with NMDA receptors. In this study, the role of NMDA receptors in the 17β-estradiol-induced pain modulation was investig...

Structure, function, and pharmacology of NMDA receptor channels.

NMDA receptors have received much attention over the last few decades, due to their role in many types of neural plasticity on the one hand, and their involvement in excitotoxicity on the other hand. There is great interest in developing clinically relevant NMDA receptor antagonists that would block excitotoxic NMDA receptor activation, without interfering with NMDA receptor function needed for...

Glutamate Receptors in Nucleus Accumbens Can Modulate Canabinoid-Induced Antinociception in Rat’s Basolateral Amygdala

Introduction: It has been shown that administration of WIN55,212-2, a cannabinoid receptor agonist, into the basolateral amygdala (BLA), dose-dependently increases the thermal latency to withdrawal in the tail-.ick test and decreases pain related behaviors in both phases of the formalin test. Recent human and animal imaging data suggest that the nucleus accumbens (NAc) is an important neural su...

Presynaptic NMDA receptor-mediated modulation of excitatory neurotransmission in the mouse dorsal motor nucleus of the vagus.

Activity of neurons in the dorsal motor nucleus of the vagus nerve (DMV) is closely regulated by synaptic input, and regulation of that input by glutamate receptors on presynaptic terminals has been proposed. Presynaptic N-methyl-d-aspartic acid (NMDA) receptors have been identified in a number of brain regions and act to modulate neurotransmitter release, but functional presynaptic NMDA recept...
